Short-range linkage relationships, genomic organisation and sequence comparisons of a cluster of five HSP70 genes in Fugu rubripes

Twelve cosmids containing sequences resembling genes encoding members of the 70-kDa heat-shock protein family, HSP70, have been isolated from Fugu rubripes. They can be broadly divided into three groups of overlapping cosmids. Restriction analysis and sequencing of one set of five cosmids have revealed five intronless Fugu HSP70 genes spanning 42 kb, arranged in a combined head-to-head, tail-to-tail and head-to-tail orientation. The levels of DNA and amino acid identity are very high with respect to one another, and are most similar to HSP70 sequences linked to the major histocompatibility complex (MHC) region in other species. Putative heat-shock consensus elements are identified. Non-HSP70 sequences with homology to known genes have been found physically linked to this Fugu HSP70 cluster: the Drosophila melanogaster SOL gene, the Drosophila melanogaster nemo gene, the Caenorhabditis elegans T17E9.1 gene and the sequence encoding the serine protease domain. The linkage relationships described here so far bear no resemblance to those of HSP70 in other organisms. Convergence of mammalian HSP70 and MHC class I and II loci probably occurred after fish had diverged. Nuclear transport mechanisms

The term nuclear transport, refers to the movement of a large variety of macromolecules both into and out of the nucleus. Transport must be extremely selective, yet also very efficient. A single type of channel, the nuclear pore complex, mediates all movement across the nuclear envelope. Selectivity is achieved through the use of families of soluble factors that target substrates for import and export and deliver them to their appropriate intracellular destinations. We now have a fairly detailed understanding of the basic mechanisms of protein import into the nucleus. Many of these same principles can be applied to protein export and perhaps RNA export. This review will summarize the current status of what is known about various transport pathways and highlight the questions that remain to be answered.     

Letters from William Burnside to Robert Fricke: automorphic functions,and the emergence of the Burnside Problem

Two letters from William Burnside have recently been found in the Nachlass of Robert Fricke that contain instances of the Burnside Problem prior to its first publication. We present these letters as a whole to the public for the first time. We draw a picture of these two mathematicians and describe their activities leading to their correspondence. We thus gain an insight into their respective motivations, reactions, and attitudes, which may sharpen the current understanding of professional and social interactions of the mathematical community at the turn of the twentieth century. Dedicated to Heiko Harborth on the occasion of his seventieth birthday.     

Cholinergic, monoaminergic and glutamatergic changes following perinatal asphyxia in the rat

Perinatal asphyxia (PA) is considered to lead to a variety of brain disorders including spasticity, epilepsy, mental retardation, and minimal brain disorder syndromes and may form the basis for psychiatric and neurodegenerative diseases later in life. We examined markers for neuronal transmission involved in the pathomechanisms of PA and candidates as mediators for long-term sequelae. We tested tyrosine hydroxylase (TH) and the vesicular monoamine transporter (VMAT) representing the monoaminergic system, the vesicular acetylcholine transporter (VAChT), and the excitatory amino acid carrier 1 (EAAC1), a neuronal subtype of the glutamate transporter, using immunohistochemistry on brain sections of rats subjected to graded PA. Three months following the asphyxiant insult immunoreactive (IR)-TH was decreased in striatum, hippocampus, thalamus, frontal cortex, and cerebellum; IR-VMAT was increased, and IR-VAChT was decreased in striatum. IR-EAAC1 glutamate transporter was increased in frontal cortex. The cholinergic, monoaminergic, and glutamatergic changes, still observed 3 months after the asphyxiant insult, may reflect their involvement in the pathomechanisms of PA and indicate mechanisms leading to long-term complications of PA. The variable consequences on the individual markers in several brain regions may be explained by specific susceptibility of cholinergic, monoaminergic, and glutamatergic neurons to the asphyxiant insult. Germline-specific dgcr8 knockout in zebrafish using a BACK approach

Zebrafish is an important model to study developmental biology and human diseases. However, an effective approach to achieve spatial and temporal gene knockout in zebrafish has not been well established. In this study, we have developed a new approach, namely bacterial artificial chromosome-rescue-based knockout (BACK), to achieve conditional gene knockout in zebrafish using the Cre/loxP system. We have successfully deleted the DiGeorge syndrome critical region gene 8 (dgcr8) in zebrafish germ line and demonstrated that the maternal-zygotic dgcr8 (MZdgcr8) embryos exhibit MZdicer-like phenotypes with morphological defects which could be rescued by miR-430, indicating that canonical microRNAs play critical role in early development. Our findings establish that Cre/loxP-mediated tissue-specific gene knockout could be achieved using this BACK strategy and that canonical microRNAs play important roles in early embryonic development in zebrafish.     

Research and Reflection: Potential Impact on the Professional Development of Undergraduate Occupational Therapy Students

In occupational therapy education in South Africa, community service (CS) focuses learning opportunities during fieldwork placements. CS therefore enabled the researcher to utilise successive small scale research projects to guide learning of students during fieldwork while simultaneously developing the occupational therapy service at a residential care facility. This community setting provided a powerful environment through which research, in combination with opportunities for reflection, contributed to nurturing skills needed by these future health professionals. A technical action research (AR) approach incorporated AR cycles and opportunities for structured reflection. Therefore situations were created for the students to embrace experiential learning. Experiential learning in the form of anticipatory reflection, reflection-in-action, reflection on reflection and retrospective reflection impacted on the quality of the students’ work. Besides encouraging unique leaning opportunities for students when engaging in research during their fieldwork placement, engagement in AR cycles simultaneously improved service delivery to residents in the facility. Key benefits of this investigation were that fieldwork education utilising AR cycles within a CS situation promoted students to identify voids in their theoretical background as well as practice skills; to apply reflective practice that could contribute to their personal and professional development; and to utilize learning opportunities optimally. Despite positive gains showed by this study, the role of power relations between the researcher as fieldwork educator and the students prohibited the AR cycles from being more emancipatory in nature and should be addressed in follow-up studies.
